A Different Way of Approaching Drug Resistant TB

Abstract

Drugs that kill tuberculosis more quickly could shorten chemotherapy significantly. In Escherichia coli, a common mechanism of cell death by bactericidal antibiotics involves the generation of highly reactive hydroxyl radicals via the Fenton reaction. Here we show that vitamin C, a compound known to drive the Fenton reaction, sterilizes cultures of drug-susceptible and drug-resistant Mycobacterium tuberculosis, the causative agent oftuberculosis. While M. tuberculosis is highly susceptible to killing by vitamin C, other Gram-positive and Gram-negative pathogens are not. The bactericidal activity of vitamin C against M. tuberculosis is dependent on high ferrous ion levels and reactive oxygen species production, and causes a pleiotropic effect affecting several biological processes. This study enlightens the possible benefits of adding vitamin C to an anti-tuberculosis regimen and suggests that the development of drugs that generate high oxidative burst could be of great use in tuberculosis treatment.

Researchers looking for options to control multi-drug resistant tuberculosis were surprised to learn that vitamin C given along with iron, could wipe out a wide variety of strains---at least in vitro.
Reporting in Nature last month, Dr. William Jacobs, Jr., professor of microbiology, immunology & genetics at Yeshiva University, NYC, said, "Mycobacterium tuberculosis is extraordinarily sensitive to killing by a vitamin C-induced Fenton reaction."
The findings suggest that tuberculosis could be prevented or treated in populations with subclinical infections using simple and inexpensive nutritional supplements---an important discovery in a time when antibiotic-resistant strains are on the rise. This has not yet been tested in infected humans, but it's certainly plausible.
"The bactericidal activity of vitamin C against M. tuberculosis is dependent on high ferrous ion levels and reactive oxygen species production, and causes a pleiotropic effect affecting several biological processes," Dr. Jacobs and his colleagues noted.
Making a Killing
"We started our research about two years ago, when we made a surprise discovery," he Dr. Jacobs told Holistic Primary Care. "We predicted that if we added isoniazid and cysteine to isoniazid-sensitive tuberculosis in culture, the bacteria would develop resistance. We knew isoniazid acts as a reducing agent, generating reactive species, so we tried another agent, vitamin C, to replace cysteine, but instead of causing resistance, we ended up killing off the culture--- something totally unexpected."
He cautioned that scientists don't yet know whether patients vulnerable to tuberculosis should take vitamin C. "We know that for this killing to occur, you also need to take the element iron along with vitamin C; this is something that could be explored, maybe in synergy with existing drugs, since one third of the world's population is sub-clinically infected with tuberculosis."
The problem, he says, is that "it would cost a lot of money to research further, and drug companies are not inclined to do that research because they can't get a return on their investment, since vitamin C is not patentable," he explained.
Meanwhile, the researchers at Yeshiva are trying to replicate their findings in vivo. "We've done the experiment in mice just a few months ago, and it did not work," said Jacobs. "But mice metabolize vitamin C differently from humans; mice make vitamin C, whereas humans can't."
Dr. Jacobs' team has also tested Vitamin E, but it didn't kill tuberculosis."
He is hoping to continue the work on vitamin C, suggesting that the combination of ascorbic acid and iron might help boost the efficacy of drug regimens.
"Vitamin C is safe and inexpensive, so drug companies should go head and do a clinical trial, although we don't know how the vitamin C would get to where tuberculosis is, in a macrophage in the patient's lungs," he said.
No Resistance

TB doesn't appear to develop resistance to vitamin C. "In a genetics lab, we typically discover things by isolating resistant mutants. We tried to find resistant mutants to vitamin C numerous times, but we've been unsuccessful, which is exactly what you'd want in a good drug," Dr. Jacobs explained.
However surprising, these findings are not the first time a study has shown that vitamins are helpful in combating TB. "For example, vitamin D actually turns on macrophages to kill TB, but that is a different mechanism; vitamin D actually stimulates an immune response," Dr. Jacobs said. The vitamin C and iron combo has a more direct cytotoxic effect.
"We need more funds to explore this; right now is a very difficult time at the National Institutes of Health to get grants. Typically the NIH will fund 25% of grants, but because of budget cuts they're only funding 6% of grants, and typically to develop a drug for use in people would cost half a billion dollars," explained Dr. Jacobs. However, it shouldn't be nearly as expensive to research the viability of vitamin C as a therapy for TB.
"If I had the money, I would go ahead and think abut doing this as an early bacteriocidal therapy. First, I would just monitor the amount of vitamin C in a patient's blood, when we gave it to them. Before a patient newly-infected with TB started on standard therapy, I would like to do a two-week trial with vitamin C as one of agents. That way, we could test a new regimen, to see if the tuberculosis got killed or not, and then go back and start them on standard therapy."

TB for Allergies?

Just the other day Andy Weil mentioned that people with asthma usually are low in magnesium resources.  Many of us in natural health care have known this too and for a very long time.

People with asthma also have a number of other nutritional deficiencies and this can be combined with food allergy as well.

Another factor is low adrenal function and suppressed immunity associated with the 30 some vaccines children are pushed to accept from birth to teen years.  Allergy often pops up after vaccination and it also seems to increase with environmental exposure to pollutants of many kinds.

In my case I think the childhood asthma I suffered with came from the fact that both my parents were constant smokers.

If you read Natural Health News often you are aware that we do not favor vaccination.

Keeping that in mind, I have to wonder just how the idea came about to utilize TB bacteria as the base for an allergy/asthma vaccine.

I also wonder, since no longterm studies have been completed just how these developers know that if you were to take this jab, how high is your risk of contracting TB.

If you look at flu vaccine or others such as measles, meningitis, pertussis, etc., you do too frequently find outbreaks of the disease in those who have taken the jab.

'One size fits all' allergy jab for hay fever, asthma and eczema on the way

A jab that could provide a "one size fits all" approach to tackling hay fever, asthma and eczema could be available within a few years, a conference heard. 

Swiss researchers claimed allergies that blight the lives of 10 million British sufferers could be largely eradicated with a single vaccine. 
An allergy conference in London heard the “one size fits all” injection that wards off asthma, eczema, hay fever and even peanut allergies could be on the shelves within four to five years. 
Experts say if the jab, known only as CYT003-Qbg10 which has been tested on humans, is properly developed it become the “hail grail” of vaccines due to it helping ward off multiple allergies.

It would be welcome news to the estimated one in five Britons, or 10 million people, who suffer from hay fever.
 
A trial, conducted by scientists from Cytos Biotechnology, a firm based in Zurich, concluded that a course of the vaccine was almost as good as steroids at keeping asthma under control.

At the jab’s heart are pieces of synthetic DNA similar to those found in the bug that causes tuberculosis or TB.

The DNA fools the body into thinking it is under attack from a dangerous bug, kick-starting a multi-pronged immune response.

A total of 63 asthmatic patients were given the course of the jab or a series of injections of a dummy drug over two or three months.

Researchers found it cut asthma attacks or symptoms by a third.

In another trial, an injection every week over a month and half, cut the amount of runny noses and weepy eyes by almost 39 per cent.

Quality of life was boosted by 42 per cent, they added.

Dr Wolfgang Renner, the chief executive of Cytos, told the Daily Mail the results were exciting.

“We think it is a one-size-fits all mechanism,” he said.

“We are very excited about it.”

Dr Renner suggested the first large-scale human trial could start next year and a vaccine within a few years.

A spokesman for Allergy UK said: “It does sound a very promising treatment, giving hope for those with severe asthma/allergy symptoms for whom the usual treatments aren’t enough, but there is still a long way to go before it will be available.”

Leanne Metcalf, director of Research at Asthma UK, added: “Over three quarters of people with asthma also have an allergy, which can often trigger their asthma symptoms.
“We are, therefore, excited about the potential of this vaccine to make a real difference to people with asthma and allergies, especially as it has been shown in clinical trials to have relatively few side effects.”

Tuesday is thought to be the worst day of the year for hay fever.

The NHS currently estimates around ten million people suffer symptoms of hay fever – such as sneezing, runny nose and itchy eyes – in the spring and summer as grasses and trees release their pollen into the air.

But that number could reach 30 million within 20 years as city living, pollution and climate change exacerbate symptoms, experts warned earlier this year.

Vitamin D and Fatigue

It never ceases to amaze me when these multiple reports about the benfits of Vitamin D always seem to overlook the issue of deficiency directly related to faulty information promulgated since the late 70s and early 80s to cut all fat out of your diet.

If you do not have healthy saturated in your diet your diet you CANNOT absorb Vitamin D, nor can you use calcium or have good support for healthy cholesterol levels and hormone production.   


Please eat unsalted-organic butter (you can make "spread" by blending one stick of butter with 1/2 cup olive oil), and any dairy products MUST contain at least 2% butter fat.

Immune Supression for Osteoporosis?

UPDATE: August 25 - Feedback on bisphosphonate drugs which contain fluoride, crumbling bones right before your eyes.
I am a femur-fracture survivor...bilateral. The right leg broke in March, the left one in July, 2009. I also suffered a compression fracture at L1 in May.

I was given Fosamax 10 years ago as a preventive measure. Now I am worse than I would have been 20 years down the road. I cannot begin to tell you that there are women breaking a leg everyday! And they do not know why. It was through the fact, that the orthopaedic doctor who took my emergency case on the first leg, had just received some information about the bilateral fractures. If you held my x-ray up against several other womens, you could not tell us apart!

I hope you continue to write and correspond about this important subject. My life has been taken away from me in a way that I never dreamed would happen.

These two articles arrived this morning in the same newsletter. I thought it interesting because I had just posted on saturated fat and mentioned how it is necessary to help make Vitamin D work.

Then there's the article about Vitamin D that arrived earlier in the day. There must be a message here.

Of course this is madness to me because there is no consideration of what natural approaches might be more effective and less expensive that a drug that the FDA now is worried about it suppressing immunity.

There's weight bearing exercise, even if you have to so it sitting. And there's Vitamin K, another fat soluble vitamin. Nettle can help too because it is a very potent cytokine reducer (what the TNF drugs do) without suppressing your immune system.

This drug is very much like the Rheumatoid Arthritis drugs, RA is often connected to wheat, gluten and gliaden allergy. Perhaps a RAST test might be a better place to start.

Related to the issue of osteoporosis and immune suppression is this information about fluoride based antibiotics. These drugs also increase risk of tendon rupture, interfere with proper thyroid function, as well as encourage brittle and weaker bones.

$33M for TB

I have pointed to this research for many years, and I'll take the opportunity again to report this.

Garlic has proven in microbiology studies to effectively treat drug resistant TB.

It would appear to me that an inexpensive approach to this seemingly major public health problem could be promptly implemented for much less that the $33M Bill Gates just donated to China for more studies and an attempt to come up with a new drug, and some patient tracking.
